Abstract
In my seminar I will discuss a series of studies from the lab aimed at understanding the relationship between the structure of receptor-ligand complexes and signaling. The main focus will be on our efforts to engineer protein ligands, using combinatorial biology, to engage receptors in alternative fashions in order to alter cell subset specificity and/or signaling amplitude. We have found that signaling by dimeric cell surface receptors, principally cytokine receptors, is surprisingly ‘tunable’ through ligand engineering. I will also discuss some recent attempts to apply these principles in a GPCR system.
