DESCRIPTION:In a functional screen of 454 single gene knockout mice we ide
 ntified genes with previously unknown roles in the immune system\, that im
 pact T cell killing. We have mapped many different steps in the order of e
 vents that lead to secretion to allow us to pinpoint when and where mutati
 ons disrupt effector function in killer T cells. We have now characterized
  several of these mutants using a combination of imaging\, biochemical\, f
 unctional and proteomic approaches to identify the molecular mechanisms th
 at allow these novel immune genes to regulate T cell. This presentation wi
 ll focus on two mutations that revealed an unexpected role for mitochondri
 a in controlling T cell killing and the future studies emerging from these
  findings.
