DESCRIPTION:Our laboratory is blending new RNA chemistries\, largely direc
ted at the ribose 2'-hydroxyl group\, with quantitative interpretation to
create new kinds of chemical microscopes for highly accurate RNA structure
analysis. We apply these technologies to important\, and otherwise intrac
table\, problems in biology. We have developed a working model for the arc
hitecture of an entire HIV-1 genome. This secondary structure model repres
ents\, in essence\, a diverse set of hypotheses for how RNA structure gove
rns viral replication. Recent functional studies supporting many of these
structural hypotheses\, plus new work focused on understanding RNA tertiar
 y structure\, will be emphasized.
