DESCRIPTION:Although forward genetic screens have identified a number of R
NA silencing components\, Chlamydomonas reinhardtii provides an ideal syst
em to establish such screens. Due to its ease of mutagenesis\, single tiss
ue-type\, ability to survive without small RNAs and haploid nature\, a wid
er range of silencing mutants can be obtained through this system. Here\,
I present three different mutant screens established in C. reinhardtii to
identify novel components of the miRNA and chromatin silencing ways and to
elucidate how they work in C. reinhardtii. I also outline a reverse genet
ics approach to determine the mechanism of chromatin silencing in C. reinh
 ardtii
