DESCRIPTION:Proteins that sense and repair lesions in DNA maintain the int
egrity of the human genome during cell division\, preventing the genetic i
nstability that initiates diseases like cancer. These molecules work in mu
lti-protein assemblies whose dynamic formation and dispersal underlies the
reactions that lead to DNA repair\, but how these protein-protein interac
tions are regulated is not well understood. In this talk\, I will describe
our work on the interaction between the breast cancer suppressor protein
BRCA2 and its partner\, the DNA repair enzyme RAD51. We have combined meth
ods from genetics\, biochemistry and biophysics to describe how BRCA2 cont
rols the availability and polymerization of RAD51 during the repair of dou
ble-strand DNA breaks in dividing cells.
