DESCRIPTION:Late onset Alzheimer’s disease (LOAD) is the most prevalent
form of dementia\, and it has no cure at present. Cholesterol metabolism i
s closely associated with AD at several stages. Cholesterol ester (CE) is
the storage form of cholesterol. A causal relationship exists between LOAD
and increased CE content in the brain. Acyl-CoA:cholesterol acyltransfera
se 1 (ACAT1) converts free cholesterol to cholesteryl esters in various ti
ssues including the brain. Here I review the recent evidence that implicat
es ACAT1 as a potential therapeutic target for AD. I also discuss the pote
ntial usage of various ACAT inhibitors currently available to treat AD.
