DESCRIPTION:Understanding the factors that produce tissue organization dur
ing development is one of the most fundamental goals in developmental biol
ogy. Various scenarios have been proposed in order to explain cell sorting
and tissue organization on the basis of the differential properties and b
ehaviors of the constituent cells. However\, validating these scenarios ha
s been notoriously difficult due to the lack of appropriate tools needed t
o simultaneously measure the relevant parameters. Here we show that by usi
ng a combination of single cell force spectroscopy\, tissue culture and in
vivo transplantation experiments\, we are able to quantify the specific a
dhesive and mechanical properties of the different germ layer progenitors
from gastrulating zebrafish embryos. With these values in hand\, we propos
e a coherent scenario that accurately predicts germ layer organization dur
 ing zebrafish gastrulation.
